Who might be able to join this trial:

• You are willing to take part and sign a consent form before any procedures begin.
• You are between 18 and 80 years old (inclusive).
• You are generally able to carry out daily activities with little or no limitation (confirm with trial site regarding specific fitness level required).
• You are expected to live for at least 3 more months.
• You have been diagnosed with an advanced gastrointestinal (digestive system) tumour that has been confirmed by a tissue or cell sample, and standard first-line treatment has not worked for you or you were unable to tolerate it.
• Your tumour tests positive for a specific protein called CLDN18.2.
• You have at least one tumour that can be measured on a scan.
• Your blood test results show that your organs (such as liver, kidneys, and bone marrow) are working well enough, tested within 7 days before starting treatment.
• You are able to communicate clearly with the research team and are willing and able to follow the study requirements.

Who may not be able to join:

• You have a type of stomach or oesophageal cancer known to be HER2-positive.
• You have taken part in another clinical trial within the 28 days before starting this trial.
• You have received any anti-cancer treatment within 21 days before starting this trial.
• You previously had serious immune-related side effects from immunotherapy, including a serious heart inflammation (confirm with trial site for specific groups this applies to).
• You still have side effects from a previous cancer treatment that have not improved to a mild level.
• You currently have moderate or severe nerve problems affecting your feelings or movement in your hands or feet.
• You have pain from your cancer that cannot be controlled.
• You have cancer that has spread to your brain or spinal cord and is causing symptoms or is currently active.
• You have a build-up of fluid in body cavities (such as the abdomen or chest) that cannot be controlled.
• You have a known allergy or serious reaction to antibody-based treatments.
• You have previously been treated with a drug that targets the same protein as the treatment in this trial.
• You have taken certain strong medications that affect how the liver breaks down drugs, within 14 days before starting the trial (confirm with trial site for specific medications).
• You have received a live vaccine within 28 days before starting the trial.
• You have a current or past lung condition called interstitial lung disease or pneumonia that required steroid tablets or injections as part of your treatment.
• You are currently taking blood-thinning medications such as heparin or warfarin.
• You have ongoing, uncontrollable vomiting that keeps coming back.
• You have had severe, uncontrolled gastrointestinal bleeding, ulcers, or diarrhoea within the 28 days before starting the trial.
• You have had a major operation or significant medical procedure within 28 days before starting the trial.
• You have been diagnosed with another type of cancer within the past 2 years.
• You have a serious heart or cardiovascular condition.
• You have another serious illness that is not well controlled.
• You are taking steroid tablets or other medicines that suppress your immune system at a significant dose, within 2 weeks before starting the trial (confirm with trial site for specific doses).
• You have a known history of an autoimmune disease (where the body's immune system attacks itself).
• You have a history of a condition that weakens your immune system.
• You have HIV, or an active hepatitis B or hepatitis C infection.
• You are a woman who could become pregnant and have tested positive on a pregnancy test within 7 days before the first dose, or you are currently breastfeeding.
• You have a known mental health condition that may make it difficult to follow the trial requirements.
• The trial doctor has decided for any other reason that taking part would not be suitable for you.
