Who might be able to join this trial:

• You have been diagnosed with stomach cancer, cancer of the gastroesophageal junction (where the stomach meets the esophagus), or esophageal adenocarcinoma that cannot be removed by surgery or has spread to other parts of the body, confirmed by a tissue biopsy.
• You have not yet received treatment for your advanced or spreading cancer (previous treatment given before or after surgery is allowed, as long as it finished at least 6 months ago).
• Your tumor tests positive for a protein called CLDN18.2, based on lab testing of your tumor tissue.
• Your tumor tests positive for a marker called PD-L1, with a combined positive score (CPS) of 1 or higher.
• You have at least one tumor that can be measured on a scan, according to standard guidelines.
• You are generally well enough to carry out light activity and care for yourself (this is measured on a standard scale called ECOG, scores 0 or 1).
• Your blood counts, liver, and kidney are working well enough to meet specific lab value thresholds (confirm exact values with the trial site).
• You are expected to live for at least 90 days (approximately 3 months).
• If you are a woman who could become pregnant, or a man, you must be willing to use effective contraception during the study and for a set period after treatment ends.
• You are willing and able to give informed consent and follow all study procedures.

Who may not be able to join:

• Your tumor tests positive for a protein called HER2.
• You have been diagnosed with another cancer within the past 3 years (some exceptions may apply for certain skin or cervical cancers — confirm with trial site).
• You have had an active or unstable stomach or intestinal ulcer, or gastrointestinal bleeding, within the past 6 weeks.
• You have an active autoimmune disease that has needed systemic (whole-body) treatment in the past 2 years, or you are currently taking medication that suppresses your immune system.
• You currently have lung inflammation (pneumonitis), or have had it in the past 3 years requiring steroids or immune-suppressing treatment.
• You are currently taking part in another clinical trial that involves a treatment intervention.
• You have had major surgery or a serious injury within the 4 weeks before starting the trial, or you are planning major surgery within 6 months.
• You have recently received radiotherapy and have not fully recovered within the timeframes set out in the trial protocol (confirm with trial site).
• You have active cancer that has spread to the brain or the lining around the brain (previously treated and stable brain metastases may be allowed — confirm with trial site).
• You have significant heart problems, such as severe heart failure, a recent heart attack, unstable chest pain, a recent stroke or mini-stroke, or major heart procedures within the past 6 months.
• You have an active or uncontrolled HIV, hepatitis B, or hepatitis C infection, or a condition that weakens your immune system (well-controlled infections may be considered — confirm with trial site).
• You have received a live vaccine within 30 days, or any other vaccine within 7 days, before your first dose of the study treatment.
• You currently have an active infection that requires intravenous (IV) antibiotics or other IV medicines.
• You have a known allergy or sensitivity to any of the study drug ingredients, including a condition called DPD deficiency (confirm with trial site).
• You have any other health condition or abnormal lab result that the trial doctor believes could put you at risk or affect your ability to take part.
