Who might be able to join this trial:
- You are between 40 and 60 years old, of any gender or race
- You have a normal or near-normal memory and thinking score on a standard screening test (confirm with trial site)
- You are willing and able to have MRI scans
- You are willing to have PET scans that involve injections of small amounts of radioactive tracers
- You fit into one of three body/metabolism groups: (1) overweight or obese with signs of blood sugar problems, (2) overweight or obese but with healthy blood sugar levels, or (3) a healthy body weight with healthy blood sugar levels
- Your body measurements allow you to fit safely inside the PET/CT and MRI scanning machines (maximum body circumference under 165 cm)
- Your BMI falls within the range required for whichever group you qualify for (confirm with trial site)
Who may not be able to join:
- You have any condition the research team feels could put you at risk, make the procedures too difficult, or affect the results — for example, if you are currently taking medication to treat obesity
- You are planning to have weight loss (bariatric) surgery
- You are unable to lie still during scans, for example due to severe long-term back pain
- You have severe claustrophobia (fear of enclosed spaces)
- You are currently pregnant or breastfeeding
- You are currently taking part in another obesity clinical trial or taking an obesity study drug
- You have been diagnosed with Type 1 or Type 2 diabetes, or have had blood sugar readings that already indicate diabetes
- You have certain blood vessel problems in both arms that would make blood draws difficult or unsafe
- You have an active skin infection or a bruise/blood pooling at the site where blood would be drawn
- You have a history of low blood sugar episodes affecting the brain caused by prolonged fasting
- You have a medical device or implant that is not safe for use in an MRI machine
