Who might be able to join this trial:

• People between 3 months and 21 years of age
• People diagnosed with a type of brain tumour called an optic pathway glioma, confirmed either by a tissue biopsy or by a brain MRI scan combined with typical clinical signs
• People whose tumour can be measured on scans, where less than 95% of the tumour was removed by surgery, or where at least 1.5cm² of tumour remains after surgery
• People with a general health and function score of at least 50 out of 100 (measured using the KPS scale for those over 12 years old, or the Lansky scale for those 12 years old and under)
• People experiencing symptoms such as reduced vision, loss of part of their visual field, swelling of the optic disc, a protruding eye, raised pressure inside the skull, or problems related to the part of the brain called the diencephalon
• People whose major organs (such as the heart, liver, and kidneys) are functioning adequately

Who may not be able to join:

• People for whom an MRI scan is not possible
• People who are unable to complete vision tests
• People whose tumour has a specific genetic change called an H3K27 mutation, even if the tumour is classified as a lower grade (grade 1 or 2)
• People currently participating in another clinical trial involving an experimental treatment
• People with a history of serious allergic reactions to medicines that are chemically or biologically similar to the drugs used in this trial
• People who have previously received an organ transplant
• People currently infected with HIV or syphilis (treponema pallidum)
• People with serious heart conditions, including specific abnormal changes seen on a heart tracing (ECG) such as T wave or ST segment changes
• People with blood clotting disorders or who are taking blood-thinning or clot-dissolving medications; people who have had significant bleeding episodes in the 3 months before joining (such as gastrointestinal bleeding, stomach ulcers with bleeding, bowel perforation, a high level of blood detected in stool, bleeding within the tumour or brain, or inflamed blood vessels); or people who have had a blood clot event such as a stroke, mini-stroke, deep vein thrombosis, or pulmonary embolism in the 6 months before joining
• People who are pregnant or breastfeeding
• People considered unsuitable for the trial for other reasons, as determined by the treating doctor (confirm with trial site)
