Who might be able to join this trial:
- People aged 18 to 50 years old
- People with a body mass index (BMI) between 25 and 40 (a measure of body weight relative to height)
- Both men and women are eligible
- People who are not currently pregnant or breastfeeding
Who may not be able to join:
- People with Type 1 (insulin-dependent) diabetes
- People with vascular (blood vessel) disease
- People who have lost more than 10 pounds in the past 2 months
- People who exercise very heavily and frequently, such as marathon runners or heavy weightlifters
- People who have had abdominal surgery, including weight loss surgery or removal of part or all of the stomach or bowel
- People with untreated thyroid disease
- People with a neurological (brain or nerve) disease
- People with a serious medical condition that the trial doctor believes could put them at risk or affect the study results
- People with chronic (long-term) pain
- People with an active psychiatric illness (including eating disorders) as defined by the DSM IV — the condition must not have been present for at least 2 years; people who screen positive on a specific psychiatric assessment tool (called the MINI+) will not be eligible
- People currently taking medications known to affect hunger or appetite
- People who are pregnant, breastfeeding, or less than 6 months postpartum
- Women who are able to become pregnant but are not using birth control, or who plan to become pregnant during the study
- People who have used oral or intravenous (IV) antibiotics in the last 3 months
- People who have used probiotic supplements in the last 3 months
- People with heavy alcohol or drug use, as identified by a positive result on the MINI+ assessment; people who smoke more than half a pack of cigarettes per day
- People who test positive on the MINI+ for drug dependence or abuse (cannabis use alone is not automatically exclusionary)
- People who have made significant changes to their usual diet or lost more than 10 pounds in the last 2 months
- People taking any medication included on the trial's specific medication exclusion list (confirm with trial site for the full list)
- People who are not fluent in English, as the study uses validated questionnaires that are currently only available in English
- People whose body weight at enrolment is greater than 400 pounds
- For the optional MRI portion of the study only: people who cannot safely have an MRI, such as those with claustrophobia or certain medical devices like pacemakers, neurostimulators, or metallic implants
Note: People on long-term maintenance medications may still be eligible, provided the dose has been stable for at least 6 months (confirm with trial site).
