Who might be able to join this trial:
Part A (Healthy volunteers):
- Adults aged 18 to 60 years, male or female
- Body mass index (a measure of weight relative to height) between 28 and 35, which has been stable (within about 5%) for the past 3 to 6 months — or up to 40 for certain expanded groups
- Generally healthy, as determined by the trial doctor based on medical history, physical check-up, and lab tests
Part B (People with obesity and a metabolic condition):
- Adults aged 18 to 60 years, male or female
- Body mass index between 35 and 50
- Thyroid hormone levels within the normal range at the screening visit — people on stable thyroid hormone medication for at least 60 days may still be considered
- A diagnosis of pre-diabetes or type 2 diabetes
Who may not be able to join:
Part A:
- People with a history of heart disease, including heart failure, heart attack, chest pain (angina), or significantly abnormal lab results
- People with a history of thyroid conditions
- People with a history or diagnosis of liver disease
- People who have used a type of medication called an siRNA agent in the past 12 months
- People who have taken part in another clinical trial or received an experimental treatment within the past 90 days (or longer, depending on the medication involved)
Part B:
- People with a significant history of heart disease, as determined by the trial doctor
- People who have used prescription anti-obesity or psychiatric medications within the past 14 days or longer (depending on the medication) — certain blood pressure and cholesterol medications may be allowed
- People currently taking more than 2 blood pressure medications, or whose blood pressure medication dose changed in the 60 days before screening
- People currently taking more than 1 cholesterol-lowering medication, or whose cholesterol medication dose changed in the 60 days before screening
- For the pre-diabetes groups only: people who have used certain diabetes-related medications (GLP-1 receptor agonists or dual incretin agonists) in the 4 months before screening
- For the type 2 diabetes groups only: people who have used insulin or certain other diabetes medications that stimulate insulin production in the 60 days before screening (confirm with trial site for full list)
- People who have used a type of medication called an siRNA agent in the past 12 months
- People who have taken part in another clinical trial or received an experimental treatment within the past 90 days (or longer, depending on the medication involved)
