Who might be able to join this trial:

• You are 18 years of age or older
• You have been diagnosed with advanced or spreading non-squamous non-small cell lung cancer (a specific type of lung cancer), confirmed by a tissue or cell sample, and your cancer has stopped responding to a type of targeted treatment called third-generation EGFR-blocking drugs, with a specific gene change (called 19 Del or 21 L858R) confirmed by a tissue or blood test
• Your lung cancer is not the small cell type or squamous cell type, and does not have certain other specific gene changes that would make you eligible for other targeted treatments (such as HER2, KRAS, BRAF, ALK, RET, NTRK, or MET changes)
• You are reasonably active and able to care for yourself day-to-day (your doctor will assess this using a standard scale scored 0–2)
• Your doctors expect you to live for at least 12 weeks
• You are able to swallow tablets or capsules
• Your blood counts, liver, and kidney are working well enough, based on recent blood tests (your doctor will check these specific values)
• If you are a woman who could become pregnant, you must have a negative pregnancy blood test within 3 days of starting the trial and agree to use reliable contraception during the trial and for 3 months after finishing
• If you are a man with a partner who could become pregnant, you must either have had a sterilisation procedure or agree to use reliable contraception during the trial and for 3 months after finishing
• You are willing and able to follow all trial visits and procedures
• You are able to understand what the trial involves and sign a consent form

Who may not be able to join:

• You have a rare or uncommon type of EGFR gene change (confirm with trial site)
• You have previously been treated with a chemotherapy combination including pemetrexed and a platinum-based drug
• You have advanced or symptomatic cancer that has spread to the brain or the lining of the brain or spinal cord
• You have active hepatitis B, hepatitis C, HIV, or syphilis (based on blood test results)
• You are pregnant, breastfeeding, or have a positive pregnancy test within 7 days before starting treatment, or you are not willing to use effective contraception during treatment and for 3 months afterwards
• You have taken, or need to take, certain medicines that affect heart rhythm or that strongly interact with a liver enzyme called CYP3A within 14 days before or during the trial (confirm specific medicines with trial site)
• You have had major surgery or immunotherapy within the 4 weeks before joining, or radiotherapy within the 2 weeks before joining
• Scans show that the tumour is growing into or very close to major blood vessels in a way that could cause serious bleeding
• You have a history of scarring or inflammation of the lung tissue (interstitial lung disease), or this is found on your baseline scan
• You have other serious ongoing health conditions — such as uncontrolled diabetes, significant mental health conditions, or other medical problems — that your doctor believes could make the trial unsafe for you or affect the results
• Your doctor has another reason they feel makes the trial unsuitable for you
