Who might be able to join this trial:

• People who have been newly diagnosed with a type of brain cancer called glioblastoma (GBM), confirmed through a tissue sample, with a specific biological marker called IDH wild type.
• People whose brain tumor tests positive for a protein called EGFR, based on a certified laboratory test.
• For the first part of the trial only (Safety Lead-in): People whose tumor shows any type of change (alteration) in the EGFR gene.
• For the second part of the trial only (Randomized Controlled Trial): People whose tumor has a specific EGFR change known as EGFRvIII.
• For the second part of the trial only (Randomized Controlled Trial): People whose tumor has an unmethylated MGMT promoter, confirmed by a validated laboratory test (confirm with trial site).
• People who have received no treatment for their glioblastoma other than surgery, followed by the standard course of radiation (54 to 60 Gy) and chemotherapy with a drug called TMZ.
• People who finished radiation therapy at least 4 weeks before joining, and whose most recent brain scan (MRI) after radiation showed no sign of the cancer growing.

Who may not be able to join:

• People whose glioblastoma has come back, spread to multiple areas of the brain, spread outside the brain, spread to the lining of the brain or spinal cord, or spread to other parts of the body.
• People whose glioblastoma has shown signs of growing or progressing before the enrolment or screening process begins.
• People who only had a biopsy and did not have surgery to remove part or all of the tumor.
• People who have previously received or are currently receiving any treatment targeting EGFR, including specific drugs such as silevertinib, bevacizumab, certain chemotherapy, immunotherapy, experimental treatments, or implanted therapies placed directly into the tumor or surgical cavity.
• People who plan to use a device called Optune® (also known as Tumor Treating Fields or TTF) during the trial.
• People with significant other serious health conditions that are not well controlled, or who have another type of cancer.
