Who might be able to join this trial:

• People aged 18 to 69 with a reduced ability to carry out daily activities (as assessed by their doctor), or people aged 70 and older, who are not suitable for a stem cell transplant in their doctor's judgment
• People who have been newly diagnosed with a type of brain cancer called primary B-cell lymphoma of the central nervous system (PCNSL), confirmed by a biopsy or fluid sample, and who have not yet received treatment for it
• People who have at least one measurable area of disease that can be tracked during the trial
• People whose kidneys, liver, bone marrow, heart, and lungs are functioning at an adequate level as assessed by blood tests and other checks
• People who have read, understood, and signed a consent form agreeing to take part in the trial
• Women who are past menopause, cannot become pregnant, or who agree to use an approved form of contraception during the trial
• Men whose partners could become pregnant and who agree to use an approved form of contraception during the trial

Who may not be able to join:

• People who have already received treatment for PCNSL, beyond a short initial course of steroids and/or a single dose of certain chemotherapy drugs (rituximab and/or methotrexate)
• People whose lymphoma has spread to parts of the body outside the brain or spinal cord
• People who have previously been diagnosed with any other type of non-Hodgkin lymphoma at any point in their life
• People whose lymphoma is only in the eye or the lining of the brain/spinal cord, with no involvement of the brain tissue or spinal cord itself
• People who have HIV infection
• People who have or have had another cancer, unless it was completely cured by surgery or there has been no sign of the cancer for at least 5 years
• People who have a known allergy or sensitivity to any of the study medications or their ingredients
• People who have mouth sores or stomach/gut ulcers that would make it unsafe to use methotrexate
• People who have an active hepatitis B, C, or E infection confirmed by a blood test
• People who have a serious active infection
• People who have a weakened immune system due to a medical condition or a previous organ transplant
• Women who are pregnant or currently breastfeeding
• People who are not able to understand what the trial involves or make their own informed decisions about taking part
• People who may not be able to follow the trial requirements, for example due to heavy alcohol use, drug dependency, refusal of blood products, or other circumstances that would make completing the trial difficult
• People who have an employment or dependent relationship with the organisation or doctor running the trial
