Who might be able to join this trial:

• You are between 55 and 90 years old (people with a known inherited form of Alzheimer's disease in the family may not be limited by age — confirm with trial site)
• You scored less than 6 points on a standard depression screening questionnaire called the Geriatric Depression Scale
• You have a caregiver who can spend at least 10 hours per week with you and can travel with you to the study location for appointments
• Your vision and hearing are good enough (with glasses or hearing aids if needed) to complete memory and thinking tests
• You are generally in good health and expected to stay well enough to participate throughout the study
• You are not pregnant, breastfeeding, or able to become pregnant (women must be at least two years past menopause or have had sterilisation surgery)
• You are willing and able to take part in brain imaging sessions over a period of time
• You scored 4 or below on a standard test used to assess blood supply problems in the brain (confirm with trial site)
• You have completed at least 6 years of schooling, or have enough work experience to show you do not have an intellectual disability
• You can speak Mandarin fluently
• You are willing to have multiple MRI brain scans and at least two PET scans
• You agree to give blood samples for genetic analysis, including tests for genes related to Alzheimer's disease risk, and for those samples to be stored and shared
• You agree to give blood samples for biomarker testing and to have those results shared

Who may not be able to join:

• A brain MRI scan shows signs of infection, stroke, or other significant damage to the brain
• You cannot have an MRI scan for any reason, such as having a pacemaker, or any metal implants or fragments in your body, eyes, or skin
• You have had severe depression or bipolar disorder in the past year
• You have had significant behavioural problems, agitation, or symptoms of psychosis in the past 3 months that might make it hard to follow the study requirements
• You are currently taking medication for obsessive-compulsive disorder or attention deficit disorder
• You have a history of schizophrenia
• You have had problems with alcohol or drug misuse or dependence in the past 2 years
• You have a serious ongoing physical illness or unstable health condition that could make it difficult to stay in a long-term study
• You have significantly abnormal levels of vitamin B12 or thyroid function that could affect the study (low B12 will lead to exclusion)
• You are currently taking certain medications that affect the brain or mood, such as certain antidepressants, antipsychotics, regular anxiety medication, or sleeping tablets; or you are taking blood-thinning medications such as warfarin, dabigatran, rivaroxaban, or apixaban (confirm with trial site for exceptions)
• You are currently using prohibited substances (confirm with trial site for details)
• You are currently taking part in another clinical trial related to brain or mental health
