Who might be able to join this trial:
Healthy volunteers:
- You have no history of any psychiatric (mental health) illness, as confirmed by a standard screening interview.
People with schizophrenia that responds to standard treatment:
- You have been diagnosed with schizophrenia according to standard international guidelines (ICD-11).
- Your symptoms are currently well-controlled (mild or minimal) and you have not had a significant return of symptoms in the past 6 months.
- You are currently taking one of these medications: Olanzapine, Risperidone, Quetiapine, Amisulpride, Aripiprazole, or Paliperidone.
- You are taking your antipsychotic medication regularly and consistently, as measured by a standard adherence scale.
People with schizophrenia that has not responded well to standard treatment:
- You have been diagnosed with schizophrenia according to standard international guidelines (ICD-11).
- You have tried at least two different antipsychotic medications in the past, each for at least 6 weeks at an adequate dose, without sufficient improvement.
- Your current symptoms are rated as relatively severe on a standard symptom scale (confirm with trial site).
- Your ability to function in daily life is moderately affected, as measured by a standard assessment scale (confirm with trial site).
- You are currently taking one of these medications: Clozapine, Olanzapine, Risperidone, Quetiapine, Amisulpride, Aripiprazole, or Paliperidone.
- You are taking your antipsychotic medication regularly and consistently, as measured by a standard adherence scale.
Who may not be able to join:
- You are currently pregnant or breastfeeding.
- You do not have the mental capacity to fully understand and agree to take part in the study.
- You have diabetes or have previously taken diabetes medication.
- You have a metal or electronic implant in your body, or dental wires, that are not safe for a very high-strength MRI scanner.
- You have severe claustrophobia (fear of enclosed spaces) that would prevent you from having a PET or MRI scan.
- Your body size is above the comfortable limits for the MRI scanner.
- You have previously taken part in research or medical procedures involving radiation, and the total radiation exposure (including this study) would reach or exceed a certain safety level within the past 12 months (confirm with trial site).
- You have had a head injury in the past that caused you to lose consciousness for more than 5 minutes.
- You have a diagnosis of an organic brain disorder (a mental health condition caused by a physical condition affecting the brain).
- You have a diagnosis of a mental health condition caused by substance use (such as alcohol or drugs).
- You have a dependence on any substance other than nicotine or cannabis (confirm with trial site).
- You have taken part in a clinical trial testing an unlicensed or experimental drug (other than a licensed antipsychotic) within the last month.
