Who might be able to join this trial:

• You are 18 years of age or older and can understand and sign a consent form
• You have been diagnosed with a specific type of esophageal cancer called squamous cell carcinoma, confirmed by a tissue test
• Your cancer cannot be surgically removed, has come back, or has spread, AND previous treatments with platinum-based chemotherapy and immunotherapy (anti-PD-1/PD-L1) have stopped working
• Your cancer can be measured on a scan, following standard imaging guidelines
• You are relatively active and able to carry out light daily activities (confirm with trial site what this level means for you)
• You have stored tumor tissue available for testing, and possibly can provide a new tissue sample if possible
• Your blood counts are at acceptable levels without needing recent transfusions or growth factor support
• Your liver is functioning well enough, based on blood test results
• Your kidneys are functioning well enough, based on a standard calculation
• If you could become pregnant or father a child, you agree to use specific birth control methods required by the study
• You are willing and able to follow all the study rules and attend all required visits

Who may not be able to join:

• You are pregnant or currently breastfeeding
• You have a known allergy or sensitivity to the study drug or its ingredients
• You are taking certain medications that are not allowed during the study
• You have received certain cancer treatments recently and have not yet recovered from their side effects (generally within the last 2–4 weeks, depending on the treatment type)
• You have not recovered from significant side effects of a previous cancer treatment (note: ongoing hair loss or nerve-related side effects at any level may still be acceptable — confirm with trial site)
• You have previously been treated with a specific class of chemotherapy drugs called topoisomerase 1 inhibitors
• You currently have another active cancer (though a past cancer that has been fully treated with no signs of return for at least 3 years, or certain low-risk cured cancers, may be acceptable)
• You have active inflammatory bowel disease (such as ulcerative colitis or Crohn's disease), or have had a bowel perforation in the past 6 months
• You currently have a serious infection that requires antibiotic treatment
• You have HIV with a detectable level of virus in your blood, or are taking medications that may interfere with the study drug
• You have active hepatitis B or hepatitis C, confirmed by specific blood tests
• You have tested positive for HIV antibodies
• You have other medical or mental health conditions that, in the doctor's opinion, could affect your ability to participate safely or complete the study
• You have taken another experimental (unapproved) drug within the past 28 days or longer before starting this study
• There is no stored or fresh tumor tissue available for testing
• Your cancer has spread to your brain and is currently active
• Your cancer is a different type called adenocarcinoma of the esophagus or gastroesophageal junction
