Who might be able to join this trial:

• People who have been diagnosed with mantle cell lymphoma (a type of blood cancer) through laboratory testing, and who have not yet received any treatment for it.
• People who are older than 18 years of age; both males and females are eligible.
• People whose cancer is at stage II, III, or IV — with stage II patients included only if the treating doctor determines that whole-body treatment is needed.
• People who have at least one area of cancer that can be measured on scans.

• People who have at least one of the following higher-risk features of their cancer: a MIPI risk score of 4–11 (a scoring tool used in mantle cell lymphoma), a Ki67 level above 50% (a marker of how fast the cancer cells are growing), a TP53 gene abnormality, or a more aggressive form of the disease known as blastic or pleomorphic type (confirm with trial site).
• People who are not considered suitable candidates for a procedure involving transplanting their own stem cells back into their body after treatment.
• People whose blood test results meet specific minimum levels for white blood cells, infection-fighting cells (neutrophils), red blood cells (haemoglobin), and platelets, and whose liver and kidney function results fall within acceptable ranges.
• People whom the treating doctor estimates have a life expectancy of more than 12 weeks at the time of joining the trial.
• People who are willing and able to take part in all required visits, tests, and procedures throughout the study.

Who may not be able to join:

• People who have previously been treated with a type of medication known as a BTK inhibitor.
• People who have serious complications or significant active infections at the time of enrolment.
• People with uncontrolled heart or blood vessel disease, blood clotting problems, connective tissue diseases, or other serious infectious conditions.
• People who currently have an active infection — such as a bacterial, fungal, or viral infection — that requires treatment throughout the whole body.
• People who are living with HIV.
• People who have a mental health condition, or who are considered unlikely to be able to follow the study requirements consistently.
• People whom the treating doctor considers unsuitable for the trial for any other reason.
