Who might be able to join this trial:
- Adults between 18 and 55 years old.
- Healthy adults with no history of major mental health conditions, as confirmed by a licensed clinical psychologist.
- Adults diagnosed with major depressive disorder (a condition involving persistent low mood and related symptoms) who are currently experiencing symptoms, as confirmed by a licensed clinical psychologist; people who also have anxiety, stress, or trauma-related conditions alongside depression may still be considered.
- People without serious ongoing or unstable physical health conditions affecting the heart, metabolism, kidneys, liver, immune system, or skin, as determined through medical history, physical checks, blood tests, and a heart trace (ECG); people with mildly elevated blood pressure (below 130mmHg), slightly raised cholesterol (below 160mg/dl), or blood sugar levels below a certain threshold (HbA1c below 5.7%) may still be considered.
- People who have a sufficient understanding of English to give informed consent, complete all tests and procedures, and attend all required study visits.
Who may not be able to join:
- People outside the age range of 18 to 55 years.
- People with a current or past diagnosis of bipolar disorder, psychotic disorders, schizophrenia, eating disorders, obsessive-compulsive disorder, or alcohol or substance use disorders.
- People assessed by a licensed clinical psychologist or study clinician as having serious and immediate thoughts of harming themselves or others, with intent, plans, or related behaviours.
- People with a diagnosed chronic physical health condition affecting the heart, metabolism, kidneys, liver, nervous system, immune system, or skin; this includes people with a resting blood pressure of 130mmHg or higher, a body mass index of 35 or above, significantly abnormal blood test results (LDL cholesterol at or above 160mg/dl, or HbA1c at or above 5.7%), or a significantly abnormal heart trace (ECG).
- People who are currently using, or have used in the past 8 weeks, medications that affect heart function or mental health, including (but not limited to) antidepressants, antipsychotics, benzodiazepines, mood stabilisers, sedatives, sleep aids, dopamine-related drugs, stimulants, buspirone, or triptans.
- People who use tobacco in any form, including electronic cigarettes.
- Women who are pregnant, breastfeeding, or planning to become pregnant; women of childbearing age will need to have a negative pregnancy test on the day of each experimental visit.
- People who currently use or have previously used hormone replacement therapy.
- People with a known allergy to the study drugs or any of the pharmacological agents used in the trial.
