Who might be able to join this trial:

People with mild Alzheimer's disease may be eligible if they meet all of the following:

• People aged 45 to 85 years old
• People who have completed at least 3 years of schooling (elementary school level or higher)
• People whose doctors have diagnosed them with probable Alzheimer's disease based on established medical guidelines
• People whose dementia has been rated as "mild" using a standard rating scale called the CDR, with a score of 1
• People whose brain scan (a type of PET scan) shows a significant build-up of a protein called amyloid beta (Aβ) in the brain

People with mild memory problems linked to Alzheimer's (called aMCI) may be eligible if they meet all of the following:

• People aged 45 to 85 years old
• People who have completed at least 3 years of schooling (elementary school level or higher)
• People who have noticed memory difficulties that someone close to them can also confirm
• People whose memory test scores fall slightly below average for their age group, based on standard testing
• People whose overall thinking and daily functioning is mostly intact, with a CDR score of 0.5 and a general thinking test (MMSE) score of 24 or above (for those with at least a junior high school education)
• People who are still able to manage everyday tasks such as using transport, shopping, and handling money
• People who do not meet the full criteria for an Alzheimer's dementia diagnosis
• People whose brain scan (Aβ-PET) shows a significant build-up of amyloid beta protein in the brain

People joining as healthy comparison participants (normal controls) may be eligible if they meet all of the following:

• People aged 55 to 85 years old
• People who have completed at least 3 years of schooling (elementary school level or higher)
• People whose general thinking test (MMSE) score is 26 or above (for those with at least a junior high school education)
• People with a CDR score of 0 (no signs of dementia)
• People with a daily living ability score of 20 or below on a standard scale
• People whose brain scan shows no significant amyloid beta protein build-up in the brain

Who may not be able to join:

• People with a history of stroke, or whose brain MRI scan shows signs of past strokes, areas of damaged brain tissue, growths, or 5 or more small areas of bleeding in the brain
• People who have another condition affecting the brain or nervous system, such as schizophrenia, severe anxiety or depression, Parkinson's disease, brain tumours, epilepsy, traumatic brain injury, or several other neurological disorders
• People who have a condition elsewhere in the body that can affect memory and thinking, such as an underactive thyroid, low levels of folate or vitamin B12, certain infections (such as syphilis or HIV), or a history of alcohol or drug misuse
• People with a serious history of liver, kidney, or lung disease, severe anaemia, serious stomach or bowel problems, a significant heart rhythm disorder, a heart attack within the past 6 months, or cancer
• People currently taking blood-thinning medications, such as warfarin or newer anticoagulant medicines
• People who cannot have an MRI scan, for example because they have metal implants in their body
• People who have difficulty communicating or a reduced level of consciousness that would prevent them from completing the memory and thinking tests
• People who are not willing to sign the consent form for the study
