Who might be able to join this trial:
- Men and women between 40 and 70 years old
- People diagnosed with knee osteoarthritis based on established medical criteria, including knee pain plus at least 3 out of a specific set of 4 clinical signs (confirm with trial site for full details)
- People who have had knee pain on most days for at least 6 months, with pain rated at a meaningful level on a standard pain scale (at least 40mm on a 100mm scale)

Who may not be able to join:

- People with significantly reduced kidney function, liver problems (including active hepatitis B or C, or abnormal liver results), or certain blood abnormalities (confirm with trial site for specific threshold values)
- People who have used steroid medications — whether injected into a joint, injected into a muscle, or taken by mouth — within the past 4 weeks
- People with a history of knee joint surgery or a condition called Charcot joint (a type of severe joint damage)
- People with a severe inward angling of the knee greater than 30 degrees
- People who have used certain disease-modifying medications (such as hydroxychloroquine or sulfasalazine) in the past 3 months
- People with a known allergy to methotrexate (a medication used in this trial)
- Women who are pregnant or currently breastfeeding
