Who might be able to join this trial:
- Adults aged 18 or older
- People who have been diagnosed with a specific type of blood cancer called large B-cell lymphoma, confirmed by laboratory testing of tissue — this includes several subtypes such as diffuse large B-cell lymphoma (DLBCL), high-grade B-cell lymphoma, lymphoma affecting the brain or spinal cord (central nervous system lymphoma), primary mediastinal large B-cell lymphoma, or follicular lymphoma that has transformed into a more aggressive form
- People who have already received a first round of treatment that included two specific types of cancer medicines: an anti-CD20 antibody (a targeted therapy) and an anthracycline (a type of chemotherapy)
- People who meet at least one of the following higher-risk conditions: the cancer did not respond well enough after 4 cycles of first-line treatment; or the cancer came back within 12 months of achieving full remission with first-line treatment; or the cancer returned after a stem cell transplant using the patient's own cells; or the cancer has spread to the brain or spinal cord at the time of relapse — OR people whose cancer has certain high-risk genetic features, specifically a TP53 gene mutation, or a high-grade lymphoma with particular gene rearrangements involving MYC and Bcl-2 (with or without Bcl-6)
- People with a general health and activity level rated as reasonably functional (confirm with trial site what this means in practice)
- People whose treating doctor assesses them as suitable to receive high-dose chemotherapy followed by a stem cell transplant using their own cells, with CAR-T cell therapy planned afterward
- People whose liver and kidney function results fall within the ranges required by the trial (confirm specific values with trial site)
- People whose heart is pumping at least 40% of its normal capacity, as measured by a heart scan
- People with a life expectancy of at least 3 months
Who may not be able to join:
- People who have previously received any treatment that targets a protein called CD19
- People whose cancer cells have been confirmed by laboratory testing to lack the CD19 protein
- People with an active hepatitis B or hepatitis C infection, meaning the virus is currently detectable at levels above the normal range in their blood
- People with an uncontrolled infection, uncontrolled heart or brain blood vessel disease, blood clotting disorders, or connective tissue diseases
- People with a history of HIV infection
- People who are pregnant or currently breastfeeding
