Who might be able to join this trial:
- People who are male and aged 18 years or older
- People who have been diagnosed with prostate cancer (a specific type called adenocarcinoma) that has spread to other parts of the body and is no longer responding to hormone-lowering treatments
- People whose cancer has continued to grow or spread despite keeping testosterone levels very low, either through ongoing hormone therapy or surgical removal of the testicles
- People whose cancer cells show certain proteins on their surface (called PSMA and/or PSCA), confirmed by a laboratory test; a special scan called a PSMA PET scan may also be used to help confirm this
- People who have previously been treated with at least one hormone-blocking medication such as abiraterone, enzalutamide, apalutamide, or darolutamide; prior treatment with chemotherapy, PARP inhibitors, radioligand therapy, or immunotherapy is also permitted
- People with a good general level of physical functioning, rated 0 or 1 on a standard scale used by doctors (confirm with trial site)
- People whose blood tests show that the heart, lungs, kidneys, liver, and blood cell counts are within acceptable ranges set by the trial
- People who have at least one area of cancer that can be measured on scans, or bone disease that can be evaluated using standard methods used in prostate cancer trials
- People with a life expectancy of at least 12 weeks
- People who are able to understand the trial, agree to take part in writing, and are willing to provide the blood and tissue samples required
Who may not be able to join:
- People who have active cancer spread to the brain or the membranes surrounding the brain or spinal cord
- People whose prostate cancer has features of a specific fast-growing type called small-cell or neuroendocrine cancer
- People who have received a type of gene-modified cell therapy within the past 6 months, or who have had a donor (allogeneic) transplant and still need medicines to suppress the immune system
- People with an active immune system condition that requires ongoing medical treatment, or who have been taking steroid medicines above a certain dose within 7 days before a procedure called lymphodepletion (confirm with trial site)
- People with an infection that is not well controlled, including uncontrolled hepatitis B, hepatitis C, or HIV
- People with serious heart problems, including an irregular heartbeat causing symptoms, a recent heart attack, or heart failure that is not well managed
- People who still have significant side effects (rated moderate or higher) from previous cancer treatments, with some exceptions such as hair loss or stable hormone-related conditions
- People who have another type of cancer that currently requires systemic treatment
- People who have a medical, mental health, or laboratory finding that the trial doctor believes would make participation unsafe or affect how results are interpreted
