Who might be able to join this trial:

  • People who have been diagnosed with Neurofibromatosis Type 1 (NF1) — either through standard clinical diagnosis criteria or through genetic testing that confirmed an NF1 gene mutation.
  • People who have at least one plexiform neurofibroma (a type of nerve tumour associated with NF1) that is either causing symptoms or not, and meets specific size requirements — at least 3 cm at its longest point as measured by examination or standard MRI, or at least 3 mL as measured by a special volumetric MRI scan.
  • The neurofibroma must be a specific type: one that has grown along the length of a nerve (possibly involving multiple branches), or a spinal neurofibroma that spans two or more levels with a connection between them or extending outward along the nerve, or a type affecting skin thickness.
  • For Phase 2 of the trial: people aged 8 years or older.
  • The person joining, or their parent or guardian, must be willing and able to sign a written consent form.
  • All participants must be able to understand, read, and speak English.
  • For Phase 2 — patients in the "with pain" group: people who report having recently experienced pain in the area of the target tumour.
  • For Phase 2 — patients in the "without pain" group: people who report having had no recent pain in the area of the target tumour.
  • For participants aged 17 or under: a primary caregiver (such as a parent, guardian, or grandparent) who is aged 18 or older, and who is able to understand, read, and speak English, must also participate.

Who may not be able to join:

  • People with severe cognitive or behavioural difficulties that, in the opinion of the trial investigators, would make it too difficult to take part in the study procedures.
  • People who have recently been enrolled in another clinical trial to treat their plexiform neurofibromas — specifically, those who joined a trial involving a type of medication called a MEK inhibitor within the past 12 months.
  • People who have started a new pain treatment — such as a new pain medication, psychological therapy, or physical therapy — within the past 3 months before enrolling in this study.

This study is not part of the standard drug-approval phase pathway (for example an observational, device, behavioural, or registry study), so a phase success rate does not apply.
