Who might be able to join this trial:
For people with Major Depressive Disorder (MDD):
- People who have been formally diagnosed with a Major Depressive Episode as part of MDD, confirmed through a structured diagnostic interview
- People aged between 25 and 55 years
- People with moderate to severe depression symptoms, as measured by a standard depression rating scale (score above 14 on the HRSD-17)
- People who have not taken any psychiatric medications for a sufficient period before the brain scan (at least 5 half-lives of the medication)
- People who are able to safely undergo an MRI scan (for example, no metal implants or pacemakers)
- For people whose depression has not been resistant to treatment: people who have previously responded to an antidepressant medication at some point in the past
For healthy volunteers (no depression):
- People aged between 25 and 55 years
- People who are able to safely undergo an MRI scan (for example, no metal implants or pacemakers)
Who may not be able to join:
For people with Major Depressive Disorder (MDD):
- People who are pregnant or breastfeeding
- People with a medical condition that requires urgent investigation or treatment
- People with a history of drug abuse or dependence within the past 6 months, or currently
- People with a lifetime history of psychosis
- People who have taken any psychiatric medication within a sufficient period before the brain scan (within 5 half-lives of the medication)
- For people whose depression has not been resistant to treatment: people whose current depressive episode has not responded to more than 2 antidepressant treatments at an appropriate dose and duration
For healthy volunteers (no depression):
- People who are pregnant or breastfeeding
- People with a medical condition that requires urgent investigation or treatment
- People with any lifetime history of a psychiatric disorder
- People who have ever been prescribed an antidepressant medication
