Who might be able to join this trial:

  • Adults between 18 and 65 years of age (inclusive)
  • People who have a current diagnosis of Major Depressive Disorder (MDD), confirmed through a structured psychiatric interview called the MINI
  • People whose depression symptoms score at least 16 on a standard rating scale called the HAM-D17, indicating a moderate to severe level of symptoms
  • People who have been consistently taking an antidepressant (specifically an SSRI or SNRI) approved by the FDA for at least 8 weeks
  • People who are right-handed
  • People who have been living at an altitude above 4,000 feet for at least 12 weeks

Who may not be able to join:

  • People who have a psychiatric diagnosis other than MDD or an anxiety disorder, as identified by the MINI interview
  • People with a history of or current kidney disease, such as chronic kidney failure, acute kidney failure, or end-stage kidney disease
  • People who currently have colitis or diverticulitis (inflammatory conditions of the bowel)
  • People with a history of or current lung disease (except for well-controlled asthma)
  • People who currently smoke
  • People with a history of heart disease, or whose heart's electrical activity shows a specific measurement (QTc) greater than 500ms on a heart tracing (confirm with trial site)
  • People with a history of fibromyalgia or any rheumatological (joint or autoimmune) condition
  • People with a history of or current seizure disorder
  • People who are currently assessed as having a serious risk of suicide, based on a standard rating scale
  • People who are currently taking antipsychotics, mood stabilisers, or antidepressants other than SSRIs or SNRIs — with the exception of bupropion at FDA-approved doses or trazodone up to 200mg — or who are taking any supplements other than standard multivitamins
  • People who are pregnant, have tested positive on a pregnancy test, or are not using an adequate method of birth control
  • People who have previously been diagnosed with serotonin syndrome, or who show signs of it currently (a potentially serious reaction to certain medications)
  • People with a pre-existing elevated level of a specific type of white blood cell called eosinophils (above 500 per microlitre of blood)
  • People who are unable to have an MRI scan, including those with certain metal implants or implanted devices, or those with severe claustrophobia (fear of enclosed spaces)
