Who might be able to join this trial:

• You are 18 years of age or older and are expected to live for at least 12 more weeks
• You have been diagnosed with a specific type of colorectal cancer (called RAS and BRAF wild-type, MSS metastatic colorectal adenocarcinoma) confirmed by a tissue or cell sample — this excludes cancers starting in the appendix or anus
• Your cancer is at an advanced stage (Stage IV) and started on the left side of the colon or rectum, and there is at least one tumour that can be measured on a scan
• You have not previously received any systemic treatments (such as chemotherapy, targeted therapies, or immunotherapy) for advanced or metastatic colorectal cancer — note: if your cancer came back during or within 6 months of completing earlier preventative chemotherapy, that treatment counts as your first treatment
• Your general health and ability to carry out daily activities is rated as good to fairly good (confirm with trial site)
• Your blood test results within 7 days before starting treatment show that your blood counts, liver function, and kidney function are within acceptable ranges (confirm with trial site)
• If you are a woman who could become pregnant, you must have a negative blood pregnancy test before starting the trial
• You are willing and able to sign a consent form, follow the study plan, and not join any other clinical trials at the same time

Who may not be able to join:

• You currently have another active cancer, unless it has been in remission for 5 or more years or was a non-invasive cancer successfully treated
• You have an active gastrointestinal condition such as a duodenal ulcer, ulcerative colitis, or bowel blockage that could increase the risk of bleeding or a bowel perforation
• You have had a stroke, mini-stroke, blood clot in the lungs, or a deep vein blood clot within the past 12 months
• You have had a heart attack, severe chest pain, significant heart failure, or serious heart rhythm problems within the past 12 months
• You have taken antibiotics for 7 or more days within the past 4 weeks, or you have had an unexplained fever above 38.5°C recently (unless the fever is linked to the tumour itself)
• You have uncontrolled fluid build-up around your lungs, abdomen, or heart in the 14 days before joining, despite receiving treatment for it
• You still have significant unresolved side effects (Grade 2 or higher) from previous cancer treatments, apart from anaemia, hair loss, or skin discolouration
• You have a lung condition such as interstitial lung disease, non-infectious pneumonia, or pulmonary fibrosis, or you have uncontrolled diabetes or high blood pressure
• You are known to have HIV, or you have active hepatitis B or C, or both
• You have a known or suspected allergy to any of the drugs used in this trial
• You are currently pregnant or breastfeeding
• You are a woman who has had a period within the last 2 years and does not use or is unwilling to use effective non-hormonal contraception, or you are a man who is able to father children (confirm with trial site)
• You have other significant physical or mental health conditions or abnormal test results that the trial team considers would make participation unsafe or unsuitable
