Who might be able to join this trial:

  • You have been diagnosed with a solid cancer that has spread or cannot be surgically removed, and your cancer has not responded to or has gotten worse after standard treatments (such as chemotherapy, immunotherapy, radiation, or targeted therapy), or no standard treatment is available or you were unable to tolerate it.
  • For the first phase of the trial: you have mesothelioma (with or without a specific gene change called NF2), OR an advanced solid tumor with an NF2 gene mutation, OR an advanced solid tumor with certain other gene changes in a pathway called the "Hippo pathway" (such as FAT1, LATS1/2, YAP, or WWTR1-CAMTA1).
  • For the second phase of the trial: you fit into one of four specific groups based on your tumor type and gene changes — details of which group applies to you should be confirmed with the trial site.
  • A specific gene change in your tumor has been identified through a genetic test (such as Next Generation Sequencing or a local lab test).
  • You are able to provide either a stored sample of your tumor tissue, or you agree to have a new tumor biopsy taken at the start of the trial.
  • Your cancer can be measured on scans according to a standard set of measurement rules (confirm with trial site).
  • You are generally well enough to carry out light daily activities — rated 0 or 1 on a standard health scale used by doctors.
  • Your blood tests show your bone marrow, kidneys, liver, and blood clotting are all working well enough.

Who may not be able to join:

  • You have cancer that has spread to the brain or spinal cord and is causing symptoms, or you have an untreated spinal cord compression.
  • You have a serious heart condition or abnormal results on heart tests.
  • You have a pre-existing or inherited kidney condition that runs in your family.
  • You are currently taking certain heart medications that affect the heart's electrical activity in a specific way (called QTc prolongation) (confirm with trial site).
  • You are currently taking certain medications that strongly affect how the body breaks down drugs through the liver — specifically ones affecting pathways known as CYP3A4 or CYP1A2 — within a certain time period before starting the trial (confirm with trial site).
  • You are currently taking certain other medications that are processed through specific liver or transport pathways in the body, within a set time before starting the trial (confirm with trial site).
  • You are currently taking certain medications that interact with specific drug transport proteins in the body (confirm with trial site).
  • You currently have a significant active infection, whether bacterial, fungal, or viral.
