Who might be able to join this trial:
- People aged 18 or older, of any gender
- People diagnosed with stage II or stage III rectal cancer who have been recommended to receive total neoadjuvant therapy (a full course of treatment given before surgery)
- People for whom a biopsy tissue sample (preserved in a standard laboratory format) is available from the time of diagnosis
- People who have given their consent to take part in the study
Who may not be able to join:
- People with stage I rectal cancer, or cancer that has come back or spread to other parts of the body
- People whose rectal cancer has spread from another cancer elsewhere in the body
- People who have been recommended for chemotherapy combined with radiation therapy only, rather than the full pre-surgery treatment program
- People who are currently being treated for cancer, or who have previously been treated for cancer
- People who do not agree to participate in the study

Primary endpoints
To investigate the detection rate and change of ctDNA in blood samples of rectal cancer patients during total neoadjuvant therapy; To investigate the relationship between ctDNA expression and MRI/CT scan imaging with pCR response in total neoadjuvant therapy (TNT); Evaluate the relationship between post-TNT ctDNA expression and disease-free survival in colorectal cancer patients in 2 years
