Who might be able to join this trial:

• Adults aged 18 to 75 years (people aged 70–75 will be reviewed by a medical committee before being accepted)
• People diagnosed with a specific type of bowel cancer (adenocarcinoma) located in the colon, sigmoid colon, or the junction between the rectum and sigmoid colon, that has grown into or through the outer lining of the bowel wall, as confirmed by a CT scan or MRI

• People whose cancer has either not spread to nearby lymph nodes, or has spread only to lymph nodes that can be surgically removed, as confirmed by CT scan or MRI
• People whose cancer has not spread to other parts of the body (no distant metastases)
• People who are fully active or have only minor restrictions on physical activity (as assessed by a standard medical scale)
• People whose cancer has a specific genetic profile known as microsatellite stable (pMMR) — this is determined by laboratory testing
• People who have given their written informed consent to take part

Who may not be able to join:

• People whose cancer has spread to distant parts of the body, such as the liver or the lining of the abdomen (metastases)
• People whose cancer is considered unable to be fully removed by surgery, as determined by a specialist medical team
• People whose cancer has a different genetic profile known as microsatellite unstable (dMMR)
• People with a deficiency of an enzyme called DPD, which affects how certain chemotherapy medicines are broken down in the body
• People who have needed emergency surgery to remove the tumour due to a blockage or perforation of the bowel (certain temporary procedures that do not remove the tumour may still be acceptable — confirm with trial site)
• People with rectal cancer located in the lower or middle part of the rectum (outside the abdominal cavity)
• People who have another serious cancer at the same time, with some exceptions for certain bowel tumours that may be discussed by the medical committee
• People with severely reduced liver, kidney, or heart function
• People who are unable to tolerate the study treatment
• People who are pregnant or breastfeeding
