Who might be able to join this trial:

• Adults aged 18 or older
• People who are well enough to carry out daily activities with little to no limitation (assessed using a standard scale called ECOG, scores 0–1)
• People who have been diagnosed with colorectal cancer (bowel cancer) that has spread to other parts of the body, confirmed by a tissue biopsy
• People whose cancer has a specific genetic change (mutation) in either the KRAS or NRAS gene
• People whose cancer has already been treated with at least 2 rounds of chemotherapy combined with drugs that block blood vessel growth, and whose cancer has continued to grow despite those treatments
• People who have previously received chemotherapy regimens containing oxaliplatin and irinotecan, and whose cancer has stopped responding to those drugs
• People who are willing and able to provide written consent to participate

Who may not be able to join:

• People with serious heart conditions, including unstable or severe coronary artery disease, a previous heart attack, advanced heart failure (classified as New York Heart Association Class III or IV), or significant irregular heart rhythms
• People who have had a stroke or a brief stroke-like episode (transient ischaemic attack) within the 6 months before screening
• People with high blood pressure that is not well controlled by medication
• People with a history of another cancer, unless it was a non-melanoma skin cancer or an early-stage cervical or breast cancer that went into full remission at least 2 years ago and requires no further treatment during the study (confirm with trial site)
• People with cancer that has spread to the brain or spinal cord, unless those areas have already been adequately treated (confirm with trial site)
• People whose blood test results fall below certain thresholds, including low white blood cell counts, low platelet counts, or low haemoglobin (anaemia)
• People with certain liver function blood test results above acceptable levels, including elevated bilirubin, ALT, or AST — though people with a condition called Gilbert syndrome may still be eligible if their results meet specific criteria (confirm with trial site)
• People with kidney function blood test results above an acceptable level
• People with a history of blood clots in veins or arteries (thromboembolic events)
• People who have had a known allergic reaction to any of the medications used in this trial
• People currently taking medications known to cause a specific type of heart rhythm problem (QT prolongation or Torsades de Pointes)
• People currently receiving any other cancer treatment, including chemotherapy, targeted therapy, or radiation therapy
• People who are pregnant or breastfeeding
• People who still have unresolved side effects of moderate severity or higher from previous cancer treatments (with some exceptions, such as mild nerve-related side effects or hair loss)
• People with any other serious medical condition, active infection, significant mental health condition, or other issue that the trial doctor believes would make participation unsafe or interfere with the trial results
