Who might be able to join this trial:
- People aged 18 or older who have given their informed consent to participate.
- People with a performance status rating of 0 or 1 on the ECOG scale, meaning they are fully active or have only minor restrictions on physical activity.
- People who have a confirmed locally advanced or metastatic solid tumour (cancer that has spread) with a documented mutation in one of the RAS genes (KRAS, HRAS, or NRAS).
- People with a solid tumour who have already received standard treatment but whose cancer progressed or who could not tolerate that treatment (for the dose exploration phase of the trial).
- People with a specific type of non-small cell lung cancer (non-squamous, without certain other gene mutations) who have not yet received systemic treatment (for specific arms of the dose expansion phase).
- People with a solid tumour or colorectal cancer who have received no more than 2 prior treatment courses for advanced disease and whose cancer progressed or who were intolerant to prior standard therapies (for another arm of the dose expansion phase).
- People whose cancer can be measured by standard imaging criteria (RECIST v1.1).
- People with adequately functioning bone marrow, liver, kidneys, blood clotting, and hormone systems.
- People who are able to swallow and take oral (by mouth) medications.
Who may not be able to join:
- People with a type of cancer called head and neck squamous cell carcinoma.
- People with any condition that might affect the ability to take or absorb the study medication.
- People who have had major surgery within the 4 weeks before starting the study drug.
- People who are unable or unwilling to attend the required study visits or complete required procedures.
