Who might be able to join this trial:

  • You are between 18 and 75 years old (any gender)
  • You have been diagnosed with Stage II or Stage III rectal cancer, with no spread to distant parts of the body, as confirmed by MRI or internal ultrasound scan
  • Your tumor is located in the lower part of the rectum, no more than 10 cm from the opening of the back passage, confirmed by a camera test or physical examination
  • Your cancer has been confirmed by a tissue sample (biopsy) to be a specific type called rectal adenocarcinoma
  • You are generally able to carry out daily activities with little or no restriction (confirm with trial site)
  • Your blood counts are within acceptable ranges, including enough red blood cells, white blood cells, and platelets
  • Your kidney function blood tests are within the normal range
  • Your liver function blood tests are within acceptable ranges
  • Your urine protein test is normal, and your blood clotting is normal
  • You have no known allergy to platinum-based chemotherapy drugs (a type of cancer medication)
  • You have not had surgery (other than a colostomy bag procedure), chemotherapy, or any other cancer treatment since your diagnosis
  • You have not previously had radiotherapy to the area of the body that would be treated in this trial
  • You are willing and able to sign a consent form agreeing to take part

Who may not be able to join:

  • You have previously been treated with certain types of immunotherapy drugs (including anti-PD-1, anti-PD-L1, or anti-CTLA-4 treatments, or other experimental immune-based therapies)
  • You have a serious autoimmune condition, such as Crohn's disease, ulcerative colitis, rheumatoid arthritis, lupus, or a similar immune system disorder
  • You have a lung condition causing symptoms, or currently have an active lung infection or inflammation
  • You have, or are at risk of, a bowel perforation (a hole or tear in the intestine), for example due to a bowel blockage or abdominal abscess
  • You have a history of another cancer, unless it was a type of non-melanoma skin cancer or a very early-stage cervical change that has been fully treated and cured
  • You currently have an active serious infection, heart failure, have had a heart attack in the last 6 months, or have unstable heart problems
  • You have other health conditions or test results that the trial doctor believes could interfere with the study or put you at greater risk
  • You are currently pregnant or breastfeeding
  • You have a condition that weakens your immune system, including HIV, or have previously had an organ transplant or stem cell transplant
  • You have an active hepatitis B infection with a high virus level, active hepatitis C infection, or active tuberculosis
  • You have received a cancer vaccine or certain other vaccines within 4 weeks before starting treatment (note: standard injected flu vaccines are generally allowed, but nasal spray flu vaccines are not — confirm with trial site)
  • You are currently taking other immune-affecting medications, chemotherapy, drugs as part of another clinical trial, or long-term steroid treatment
  • You have a mental health condition, substance misuse issue, or other personal circumstances that a doctor believes may make it difficult for you to follow the trial requirements
  • You have a known allergy or medical reason why you cannot take the drugs used in this trial
