Who might be able to join this trial:
- Adults between 18 and 70 years old (male or female)
- People with a confirmed diagnosis of rectal cancer (a specific type called adenocarcinoma) where the tumour is located in the lower part of the rectum, within about 12 cm of the anal opening
- People whose cancer has been staged by MRI scan as having grown significantly into or beyond the rectal wall (T3–T4), or where cancer has spread to nearby lymph nodes
- People whose blood counts and liver and kidney function test results fall within acceptable ranges as determined by the trial site
- People who are generally able to carry out daily activities with no or only minor limitations (confirm with trial site regarding how this is assessed)
Who may not be able to join:
- People whose cancer has spread to distant parts of the body (Stage IV/metastatic disease)
- People whose rectal cancer has come back after previous treatment
- People who are currently experiencing serious complications such as active bleeding, a bowel perforation, or other urgent conditions that require emergency surgery
- People who have already received systemic cancer treatment (such as chemotherapy) for their rectal cancer
- People who currently have another active cancer that is not a colorectal cancer
- People who have an active autoimmune disease, or a history of autoimmune disease that required steroids or immune-modifying treatments
- People who have certain lung conditions, such as interstitial lung disease or non-infectious pneumonitis, or poorly controlled conditions such as diabetes, high blood pressure, pulmonary fibrosis, or acute lung inflammation
- People who have ongoing significant side effects (grade 2 or higher) from a previous cancer treatment, with the exception of anaemia, hair loss, or skin darkening
- People who have previously been treated with certain types of immunotherapy drugs that target PD-1, PD-L1, or CTLA-4
- Women who are pregnant or breastfeeding
- People who are known to be HIV-positive or have AIDS
- People with a known or suspected allergy to any of the medications used in this trial
