Who may be able to join
- A pathology test has confirmed a diagnosis of non-small cell lung cancer (NSCLC) that cannot be surgically removed and has either spread to nearby areas or to other parts of the body.
- The cancer has at least one measurable area of disease, as determined by a standard measurement system called RECIST 1.1.
- Testing on tumour tissue has confirmed a specific genetic change in the cancer called a KRAS G12D mutation, and tissue is available for further confirmatory testing at a central laboratory.
- The person has a good general level of physical functioning, rated 0 or 1 on a standard scale used by doctors (meaning fully active or able to carry out light activity).
- The person's organs (such as kidneys, liver, and bone marrow) are functioning at an adequate level.
- The person has previously been treated with both a platinum-based chemotherapy and an immune checkpoint inhibitor for advanced or unresectable disease.
- The cancer has been documented to have gotten worse during or after the most recent treatment received.
- For people being considered for the 2nd or 3rd line of treatment (without brain involvement), on a stable or preferred dose: at least 1 but no more than 2 prior treatment regimens have been received for advanced or unresectable NSCLC.
- For people being considered for the 2nd to 4th line of treatment who have cancer that has spread to the brain: at least 1 but no more than 3 prior treatment regimens have been received for advanced or unresectable NSCLC.
- For people with brain metastases: the brain metastases are not causing symptoms and have not yet been treated.
- For people with brain metastases: at least one untreated, measurable brain lesion meets specific size requirements (longest measurement at least 0.5 cm and no more than 3 cm) under a specific measurement system (confirm with trial site).
Who may not be able to join
Each point below is a reason the trial team may not be able to accept someone. It is not a list of requirements to meet.
- The person has another documented mutation in a group of genes related to RAS (other than the KRAS G12D mutation being studied).
- The person has received any anti-cancer treatment within 4 weeks before the first dose of the trial treatment, or within a drug-specific timeframe — whichever is shorter.
- The person has had major surgery within 4 weeks before the first dose of the trial treatment.
- The person has received radiation therapy within 1 week before the first dose of the trial treatment.
- The person has a history of lung inflammation or scarring caused by a medication (known as drug-induced interstitial lung disease).
- The person has previously received a treatment that directly targets the RAS protein.
- The person has cancer that has spread to the brain and is either causing symptoms or has not yet been treated (note: people with untreated brain metastases may be eligible under the specific brain metastases group described above — confirm with trial site).
- The person has taken certain medications that strongly affect how the body processes drugs (called strong CYP3A4 inhibitors, inducers, or sensitive substrates with a narrow therapeutic index) within 14 days before the first dose, or within a drug-specific timeframe — whichever is shorter (confirm with trial site).
- The person has taken a proton pump inhibitor (a common type of heartburn or acid reflux medication) or an H2 blocker (another type of acid-reducing medication) within 5 days before the first dose.
- The person is unable to swallow oral (by mouth) medication.
- There may be additional criteria not listed here that the trial site will assess.
