Who might be able to join this trial:
- People aged 18 to 85 years old
- People who meet at least one of the following: they are a known carrier of a specific genetic mutation (in the PRNP gene); they have a biological parent and other family members who have had a prion disease diagnosis; or they are a family member who does not carry the mutation, or an unrelated volunteer who previously enrolled as a control
- People who are healthy enough to have a blood draw, a lumbar puncture (spinal fluid sample), and cognitive (thinking and memory) testing
- People with adequate vision and hearing to complete cognitive testing
- People who are fluent in English
- People who have had at least 5 years of formal education
- People who are able to understand and agree to what the study involves and can follow the study procedures
- People who have no medical reasons preventing them from having an MRI scan — this requirement applies to PRNP mutation carriers only
Who may not be able to join:
- People who have a brain or nervous system condition other than asymptomatic (no symptoms) or early prion disease — for example, stroke, brain tumour, multiple sclerosis, serious head injury with lasting effects, Alzheimer's disease, Parkinson's disease, or other known conditions that cause progressive brain deterioration
- People who have had a problem with alcohol or other substance misuse or dependence within the past two years
- People who have a significant ongoing illness, an unstable health condition, or who are pregnant, where participation could pose a safety risk or affect their ability to take part
- People who have a blood clotting disorder or who are taking blood-thinning medications (such as Coumadin/warfarin) that raise the risk of complications during blood draws or spinal fluid collection, where certain blood test results fall outside an acceptable range (confirm with trial site)
